小麦条锈菌对粉宁抗药性的敏感性研究

西南科技大学硕士研究生学位论文 第1页 摘 要 本研究采用拌种离体叶段法对四川和云南两省的84个条锈菌株 其中四
川的梓潼10个，三台8个松潘10个，苍溪2个，广元2个，盐亭10个，黑
水10个，郫县10个，德昌10个和云南12个 进行抗药性监测。依据监测结
果筛选出本研究所需的敏感性和抗药性菌株，菌株的EC50值小于50mg／l的
为敏感菌株，菌株的EC50值大于800rag／l的为抗性菌株，从中筛选出敏感菌
株2个，抗药性菌株18个；大部分菌株属于中抗菌株；供试菌株中属于敏
感菌株占2．4％，耐药 抗药性 菌株占61．9％；供试菌株的平均EC50值为
3．394mg／1，平均抗性水平为21．43倍，平均抗性频率在10～50倍的菌株占
52．35％，云南菌株的平均抗性水平高于四川菌株，同时在用一个省的不同地
区之间，菌株的EC50值也存在差异，即使是同一个地区的不同菌株之间，EC50
值也不同。因此了解小麦条锈病大范围的流行趋势是非常必要的，本研究对
小麦条锈病抗药性持续治理措施的制定以及杀菌剂生产和使用策略提供理论
依据。 根据以上监测结果筛选出来的敏感性与抗药性菌株20个，对菌株进行基
因组DNA提取，再通过等位基因特异PCR AS．PCR 技术对供试菌株群体
果发现粉锈宁的抗性不只是由单个位点基因的突变引起的。通过对不同菌株
敏感性与突变位点的关系可发现，敏感性菌株CYP5l基因上136位点未发生
突变，而抗性菌株CYP51基因上136位点则发生了突变，所以初步认定位点突
变与抗性有关。因此可将菌株的敏感性分为敏感、中抗和高抗三个梯度 敏
感 EC50≤5 、中抗 5 EC50≤50 和高抗 EC50 50 。 关键词：小麦条锈菌粉锈宁抗药性CYP51 西南科技大学硕士研究生学位论文 第1I页 Abst
